Ligation gave me a headache
its been almost 6weeks for now.... we've been trying out different approaches to move onto the my next phrase of research. We hit a few high walls already. Currently, my boss seems to be banking all our efforts onto this ONE particular series of techniques. Despite repeatedly failing to arrive at a decent conclusion, he finds condolences at the fact that we're "repeating" the protocol to refine our skills. Even though we have absolutely NO progress from week ONE.
After working around 30over sets of samples and repetition, I've kinda concluded that my biggest challenge is in Ligation. That is to physically "stick" two ends of DNA strands together. It seems like an impossible task to STICK genomic DNA to synthetic DNA. Does DNA also practise "racism"?
Maybe its the lousy ligase (enzyme) that I'm using, someone was telling me that Promega's stuff has poor QC, I guess if I can lay my hands on other ligase and maybe even some new polymerase (another enzyme) then I'll be able to verify that statement.
And until that time comes, everyday is a repetition of protocol on some new samples by some strange reason or logic is suppose to work with more practise.... headache!
Or is this God's way of telling me to give up? hee... pray for me brothers n sisters... I'll need any kind of divine interventions here....
